By bus, by car or by train:
Take the vaporetto (waterbus) n° 01 along the Canal Grande and get off at the Rialto Stop. Go straight ahead into Calle Dei Fabbri. You will find the Hotel Alcyone along the street on the left.
From the airport, the easiest solution is to take a private water taxi, it costs around 90 Euro and takes 30 minutes. The hotel has its own stop.
Otherwise You can cathch the bus n° 05 and then the waterbus as above.
You can also take a public boat called Alilaguna to Saint Mark Square. From there You should find on the left of the clock tower Calle Dei Fabbri. In this case the Hotel Alcyone is on the right.
